I turned engine by hand, put engine in the car with tranny, now I can't turn the flywheel, what could this mean?

Its not in gear, I just finished my engine swap, and today I was trying to rotate the thing to finish tighing the bolts on the flywheel. I orginally installed it to the block before returning the engine cradle and all into the car.
But now I can't turn the thing. Tranny is in netural, tried park too.

It doesn't matter if it is in a gear or not on an automatic. The torque converter will spin freely whether or not the transmission is in a gear or in neutral. I'm asuming you alredy have the flexplate bolted to the rear of the crank. Are you trying to tighten the 3 torque converter bolts that attach to the flexplate? Why didn't you do that when it was on the engine stand?
You probably just have the crank right at a compression stroke. You probably just need a little more leverage.
